BIOACTIVE COMPOUNDS OF SEA BUCKTHORNS (HIPPOPHAE RHAMNOIDES) BERRIES AND LEAVES - EFFECTS OF DRYING AND EXTRACTION METHODS

Abstract:
Berries and leaves of sea buckthorn were dried in artificial accelerated solar-drier and moisture content was measured during the process. Initial moisture of berries was found up to 82% and of leaves up to 65%. Water absorption index (WAI) and oil absorption index (OAI) of the dried products were also measured and found that it ranges from 3.26 to 5.85 WAI and 1.59 to 3.26 OAI. Sequential oil extractions were carried out on dried berries and leaves using solvents of different polarity (hexane, methanol, ethyl acetate, butanol, water). A green extraction method, the solvent-free microwave assisted extraction, was applied to fresh berries and compared to solvent extraction. The extracts were analyzed using Liquid Chromatography-Diode Array-Mass Spectrometry (LC-DAD-MS) in order to investigate their major bioactive constituents. The antioxidant activity of extracts was measured using the 2,2-diphenyl-1-picryl-hydrazil (DPPH) free radical scavenging method. The results indicated that the different extraction techniques affected the examined properties as well as the profile of bioactive compounds. Drying kinetics of the plant were examined during the different drying processes. The structure of the products was evaluated through Scanning Electron Microscopy (SEM) analysis.
