TEMPERATURE, PH AND DEVELOPMENT PERIOD ON IN VITRO POLLEN GERMINATION IN PYRUS CALLERYANA

Abstract:
Three experiments were conducted with the objective of establishing the best pH value, the ideal temperature for pollen germination and pollen tube growth in ‘Taiwan Mamenashi’ and ‘Taiwan Nashi-C’ (Pyrus calleryana Decne). For this study, the pollen grains were placed on Petri plates with 20 ml of culture medium containing 90 g.L-1 of sucrose, 10 g.L-1 of agar and 1200 mg.L-1 of boric acid. Different pH levels (4.0, 4.5, 5.0, 5.5, 6.0 and 6.5), temperature (20, 25, 30 and 35°C) and pollen tube development periods (1, 2, 3, 4, 5, 6 and 12 hours after inoculation) were tested. It was observed that, for both cultivars, pH between 5.5 and 6.0 gave the best results on germination percentages. The regression analysis showed that, after 12 hours in B.O.D., the best temperature for pollen germination was 28°C for ‘Taiwan Mamenashi’ and ‘Taiwan Nashi-C’. The pollen tube development began approximately 60 minutes after inoculation. It was also observed that the pollen tubes stopped their development after six or twelve hours in culture medium, respectively for ‘Taiwan Nashi-C’ and ‘Taiwan Mamenashi’.
