IN VITRO SELECTION FOR FUSARIUM RESISTANT ORIENTAL LILY MUTANTS USING CULTURE FILTRATE OF THE FUNGAL AGENT

Abstract:
In this study, callus cultures derived from in vitro scales of Oriental lily ‘Casa Blanca’ susceptible to Fusarium oxysporum f. sp. lilii were successfully used for in vitro selection for developing resistance to this pathogenic fungus. The suitable induction callus medium is a MS supplemented with a hormone combination of TDZ 0.05 mg L-1 and Picloram 2 mg L-1. The callus was added to different concentrations of culture filtrate of F. oxysporum to determine appropriate culture filtrate concentration to be used for disease-resistant selection. And this stock is feasible as selective agent for screening for F. oxysporum wilt resistance. Resistant cell lines were selected by culturing calli on MS medium supplemented with growth regulators and containing 80% concentrations of culture filtrate of F. oxysporum. 7.0% calli regenerated shoots on MS medium supplemented with different concentrations of naphthalene acetic acid (NAA) and benzyladenine (BA). Morphologically, the selected regenerants could be grouped in two kinds of mutants. It is certificated the selected regenerants were mutated through measuring leaf length, leaf width, leaf index and chromosome analysis. The selected clones were moderate resistant to Fusarium bulb rot.
